水溶性及固定化铬氨酸酶去除苯酚的试验研究

Preliminary Study on Removal of Phenol from Aqueous Solution by Soluble and Immobilized Tyrosinase

  • 摘要: 在考察水溶性酪氨酸酶去除配水苯酚的方法及影响酚去除率的因素基础上,采用海藻酸钙凝胶包埋一戊二醛交联结合法制成固定化酶,进行固定化酶法去除水溶液中苯酚的实验研究.结果表明,经4小时处理试验后,溶解性酶法和固定化酶法的苯酚去除率分别为72.3%和96.0%.本法制成的固定化酶的酶包埋率为75.7%,在六周内重复使用,苯酚去除率仅下降3%.水溶性酶催化苯酚的反应产物对酶活性有抑制作用,使用固定化酶可改善反应的微环境,有利于提高酚的去除率.

     

    Abstract: An enzymatic method for removal of phenol from model wastewater catalyzed by tyrosinase was studied.Based on investigation of effects on the removal efficiency of phenol using soluble tyrosinase,experiments on removal of phenol were carried out using immobilized tyrosinase,which is obtained by combining entrapment of calcium alginate with crosslinking of glutaraldhyde.The results showed that the removal efficiency of phenol after 4h by treatment with the soluble and immobilized enzyme was 72.3% and 96.0% respectively.The estimated amount of entrapped enzyme was 75.7% of added suluble enzyme,and the immobilized enzyme was repeatedly used for 6 weeks with a decrease of re moval rate by only 3%.It was inferred that products from the enzymatical reaction catalyzed by soluble enzyme could inhibit the enzyme activity.However,reactive micro-environment may be improved by using immobilized enzyme and favourable to heighten the removal efficiency.
